Description : The protein encoded by this gene is a DNA polymerase involved in base excision and repair, also called gap-filling DNA synthesis. The encoded protein, acting as a monomer, is normally found in the cytoplasm, but it translocates to the nucleus upon DNA damage. Several transcript variants of this gene exist, but the full-length nature of only one has been described to date.

Usage : Based on ProFoldin DNA Polymerase kit. DNA polymerase is incubated with dNTP mix and DNA substrate in 50 μL reaction at RT for 1 h. Detection done by addition of 200 μL reagent U (ProFoldin) and 40 μL 1× fluorescence dye. Read fluorescence at λexc=485 nm and λem=535 nm.
Notes : Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.
Storage : At least 6 months at -80 centigrade.
Storage Buffer : 40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 250 mM imidazole, 0.04% Tween-20, and 20% glycerol.

Efficacy: The enzyme is still active and I am currently using it in some DNA repair assays. It works very well for single nucleotide insertions in gapped DNA which is mostly what I have been using it for recently. Quality: Very happy with the quality and longevity. I think the enzyme is ~1yr old with frequent usage and it is still working fine. Overall Experience: Very happy. When we need more polB/hUNG, and potentially others, we will purchase from you again.
