Chitinase-3-like protein 1 (CHI3L1) shows up in high levels in gliomas and helps them grow and spread. Research has linked CHI3L1 levels to how long patients survive, and it's mostly found in glioma cells. It kicks the NF-κB pathway into gear, especially as tumors advance. CHI3L1 works with proteins like ACTN4 and NFKB1 to amp up this signaling by helping move components into the cell's nucleus. In the tumor environment, CHI3L1 latches onto CD44 on nearby macrophages, activating the AKT pathway, which turns these cells into a type that supports tumors. It also ties in with immune checkpoints such as PD-L1, making the tumor environment more immune-suppressive.

Chitinase 3-like-1 (CHI3L1) affects immune responses in cancer and COPD. It reduces adaptive immunity in cancers by activating immune checkpoints, which worsens outcomes. In COPD, it influences lung cell death. This study found that CHI3L1 in lung cancer metastasis reduces macrophage activity by activating specific checkpoints and lowering signals needed for cell processing. It also involves changes in proteins related to cell structure. By suppressing these immune responses, CHI3L1 promotes cancer and inflammation progress. Targeting CHI3L1 could offer new treatments for cancer, COPD, and similar conditions.
