• Product Overview: Thermostable recombinant RNase H is an endoribonuclease that remains active at relatively high temperatures. It can selectively recognize and cleave the phosphodiester bonds of RNA in RNA: DNA hybrid duplexes while keeping the DNA in the hybrid duplex intact. It has enzymatic properties similar to those of the common E.coli RNase H. It will not degrade single-stranded or double-stranded RNA or DNA, but it still exhibits high activity above 65 centigrade, making it convenient to be used under high-temperature conditions. • Usage: 1. Prepare the reaction system on ice: RNA: DNA complex: 2 μg 10× RNase H Reaction buffer: 10 μL Thermostable recombinant RNase H: 1 μL Water: Up to 100 μL 2. Incubate at 50 centigrade for 20 minutes. 3. Add 1 μL of 0.5 M EDTA to terminate the reaction.
• Applications: 1. Depiction of RNA structure maps with high stringency and site-specific RNA cleavage. 2. Enzymatic removal of the poly (A) of mRNA hybridized with oligo (dT). 3. Removal of mRNA after cDNA synthesis. 4. Isothermal amplification.
