Description : Tyrosine kinase that plays an essential role in regulation of the adaptive immune response. Regulates the development, function and differentiation of conventional T-cells and nonconventional NKT-cells. When antigen presenting cells (APC) activate T-cell receptor (TCR), a series of phosphorylation lead to the recruitment of ITK to the cell membrane, in the vicinity of the stimulated TCR receptor, where it is phosphorylated by LCK. Phosphorylation leads to ITK autophosphorylation and full activation. Once activated, phosphorylates PLCG1, leading to the activation of this lipase and subsequent cleavage of its substrates. In turn, the endoplasmic reticulum releases calcium in the cytoplasm and the nuclear activator of activated T-cells (NFAT) translocates into the nucleus to perform its transcriptional duty. Phosphorylates 2 essential adapter proteins: the linker for activation of T-cells/LAT protein and LCP2. Then, a large number of signaling molecules such as VAV1 are recruited and ultimately lead to lymphokine production, T-cell proliferation and differentiation. Required for TCR-mediated calcium response in gamma-delta T-cells, may also be involved in the modulation of the transcriptomic signature in the Vgamma2-positive subset of immature gamma-delta T-cells. Phosphorylates TBX21 at 'Tyr-525' and mediates its interaction with GATA3.

        Q&As (5)

        Ask a question
What is the significance of ITK in allergic and autoimmune diseases, and how does it contribute to immune dysregulation in these conditions? 01/10/2022

ITK's involvement in Th2 cell differentiation links it to allergic diseases. Dysregulation of ITK is implicated in autoimmune diseases, influencing aberrant T-cell responses and immune system dysfunction.

Can you discuss the role of ITK in the context of immunodeficiency disorders, and how genetic variations impact its function? 12/01/2021

Deficiencies in ITK function are associated with immunodeficiency disorders. Genetic variations in ITK may compromise its kinase activity, leading to impaired T-cell signaling and immune responses.

What is the primary role of the ITK protein in T-cell signaling, and how does it contribute to immune responses? 10/17/2020

ITK is a tyrosine kinase involved in T-cell signaling, specifically in the activation of T-cell receptors. It plays a critical role in the regulation of immune responses and T-cell-mediated immunity.

Can you discuss the molecular mechanisms by which ITK is activated and its downstream signaling pathways in T cells? 02/27/2019

ITK is activated by phosphorylation and participates in downstream signaling pathways, including the PLCγ1 pathway, leading to the mobilization of calcium ions and activation of NFAT transcription factors.

How does ITK participate in the regulation of immune synapse formation, and what are the implications for T-cell activation and effector functions? 09/03/2018

ITK contributes to immune synapse formation by mediating cytoskeletal changes and promoting T-cell receptor clustering. This influences T-cell activation and the subsequent effector functions.
