Description : Thy-1 membrane glycoprotein, also known as Thy-1 antigen, CD9 and THY1, is a cell membrane protein which contains 1 Ig-like V-type (immunoglobulin-like) domain. It is a glycophosphatidylinositol-linked glycoprotein expressed on the surface of neurons, thymocytes, subsets of fibroblasts, endothelial cells, mesangial cells and some hematopoietic cells. It has been identified on a variety of stem cells and at varying levels in non-lymphoid tissues such as on fibroblasts, brain cells, and activated endothelial cells. Thy-1 is evolutionarily conserved, developmentally regulated, and often has dramatic effects on cell phenotype. Thy-1 is a 25-37 kDa glycosylphosphatidylinositol (GPI)-anchored protein involved in T cell activation, neurite outgrowth, apoptosis, tumor suppression, wound healing, and fibrosis. To mediate these diverse effects, Thy-1 participates in multiple signaling cascades. Thy-1 is an important regulator of cell-cell and cell-matrix interactions, with important roles in nerve regeneration, metastasis, inflammation, and fibrosis.

The role of N6-methyladenosine (m6A) in biliary atresia (BA) is unclear. This study found increased m6A levels in BA, associated with worse liver function and fibrosis stages. MeRIP-seq and RNA-seq identified 130 m6A-modified genes linked to fibrosis. In vivo and in vitro experiments confirmed m6A's regulatory effect on COL1A1 and THY1. THY1+ myofibroblasts, which expanded in BA, expressed fibrogenic genes. THY1 was linked to liver fibrosis, jaundice clearance, and poor native liver survival in BA. Further studies showed THY1's role in extracellular matrix organization and HSC activation, suggesting its importance in BA pathogenesis.

Case 2: Zhang Q, et al. Cell Death Dis. 2018

Human glioma-associated mesenchymal stem cells (gbMSCs) play a key role in the development of malignant gliomas and are divided into CD90+ and CD90- subpopulations with distinct functions in glioma progression. This study revealed that CD90high gbMSCs enhance glioma growth by promoting cell proliferation, migration, and adhesion, while CD90low gbMSCs facilitate tumor growth by differentiating into pericytes and promoting blood vessel formation through interactions with vascular endothelial cells. Additionally, researchers identified differences in long non-coding RNAs and mRNAs expression between the subpopulations and discussed the possible molecular mechanisms involved.

THY1 Protein, also known as the CD97 antigen, is a highly conserved glycoprotein that is part of the immunoglobulin superfamily. It is expressed in a variety of cell types, including neurons, hematopoietic stem cells, activated T and B lymphocytes, neutrophils, fibroblasts, and many tumor cells.

In clinical oncology, recombinant THY1 protein can be used in molecular imaging techniques to improve the accuracy of tumor detection. For example, it can be used as a targeted ultrasound contrast agent, binding to specific ligands, to enhance tumor detection. Due to the expression pattern of THY1 in multiple cancers, recombinant THY1 proteins can be used to develop new drugs for specific types of cancer, including ovarian cancer, nasopharyngeal cancer, hepatocellular carcinoma, and others. In stem cell research, THY1 acts as a marker to help identify and isolate populations of cells with specific functions, such as mesenchymal stem cells and tumor stem cells. THY1 plays a role in skin cells, and the recombinant THY1 protein helps in the study of skin homeostasis and repair mechanisms, as well as related diseases such as skin cancer.
