Description : Plasma Selectin (P-selectin) is a cell adhesion molecule (CAM) found in granules in endothelial cells (cells lining blood vessels) and activated platelets. P-Selectin is expressed on activated endothelium and platelets where it can bind monocytes, neutrophils, stimulated T cells, and platelets. P-selectin plays an essential role in the initial recruitment of leukocytes (white blood cells) to the site of injury during inflammation. When endothelial cells are activated by molecules such as histamine or thrombin during inflammation, P-selectin moves from an internal cell location to the endothelial cell surface. Thrombin is one trigger which can stimulate endothelial-cell release of P-selectin and recent studies suggest an additional Ca2+-independent pathway involved in release of P-selectin. P-selectin-mediated adhesion is an important factor in the development of early atherosclerotic lesions.
Molecular Weight : The predicted molecular weight of Recombinant Human P-Selectin is 106 kDa. However, the actual molecular weight as observed by migration on SDS Page is 146-160 kDa under reducing conditions.
State Of Matter : Lyophilized.
Formulation : This recombinant protein was 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 – 7.3 with no calcium, magnesium, or preservatives.
Purity : >95% by SDS Page and analyzed by silver stain.
Endotoxin : <1.0 EU/µg as determined by the LAL method.
Biological Activity : Measured by its ability to bind P-Selectin ligands on U937 cells. P-Selectin/Fc immobilized at 10 μg/ml will induce greater than 50% adhesion on U937 cells (100 μl/well at 1 x 106 cells/ml).
Storage : This lyophilized protein is stable for six to twelve months when stored desiccated at -20℃ to -70℃. After aseptic reconstitution, this protein may be stored at 2℃ to 8℃ for one month or at -20℃ to -70℃ in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.
