Antigen Description : | Secretagogin, also known as SCGN, is a secreted protein which is detectable in human serum after ischemic neuronal damage. It is a recently described calcium-binding protein. Secretagogin / SCGN is expressed at high levels in the pancreatic islets of Langerhans and to a much lesser extent in the gastrointestinal tract (stomach, small intestine and colon), the adrenal medulla and cortex and the thyroid C-cells. In the brain, the expression of Secretagogin / SCGN is restricted to distinct subtypes of neurons with highest expression in the molecular layer of the cerebellum (stellate and basket cells), in the anterior part of the pituitary gland, in the thalamus, in the hypothalamus and in a subgroup of neocortical neurons. Secretagogin / SCGN is widely expressed in prostatic adenocarcinoma as opposed to adenocarcinomas in other organs. The function of Secretagogin / SCGN is unknown, but it has been suggested in beta-cells to influence calcium-influx, insulin secretion and proliferation, and has been observed downregulated in diabetes-prone BB rat islets exposed to cytokines. Secretagogin / SCGN is involved in the calcium metabolism of tumour cells and endothelial cells in a subset of neoplasms of the brain and its coverings. Secretagogin / SCGN is also a novel marker for neuroendocrine differentiation. |
Specificity : | Human SCGN / Secretagogin. No cross-reactivity with E.coli cell lysate in ELISA. |
Immunogen : | Recombinant Human SCGN / SecretagoginProtein |
Host : | Mouse |
Isotype : | Mouse IgG1 |
Species : | Human |
Clone : | 8G3C13 |
Applications : | Western blot; ELISA |
Dilution : | Western blot: This antibody can be used at 1-2 μg/mL with the appropriate secondary reagents to detect Human SCGN in WB. Using a DAB detection system, the detection limit for Human SCGN is approximately 4 ng/lane under non-reducing conditions and reducing conditions.Direct ELISA: This antibody can be used at 0.5-1 μg/mL with the appropriate secondary reagents to detect Human SCGN. The detection limit for Human SCGN is approximately 0.16 ng/well. |
Preparation : | This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, human cell-derived, recombinant Human SCGN / Secretagogin. The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography. |
Format : | 0.2 μm filtered solution in PBS with 5% trehalose |
Storage : | This antibody can be stored at 2-8°C for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20 to -70 °C. Preservative-Free.Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles. |
