Antigen Description : | Complement factor H, also known as H factor 1, and CFH, is a secreted protein which contains 20 Sushi ( CCP / SCR ) domains. It is a large soluble glycoprotein that circulates in human plasma (at a concentration of 500–800 micrograms per milliliter). The main job of CFH is to regulate the Alternative Pathway of the complement system, ensuring that the complement system is directed towards pathogens and does not damage host tissue. CFH regulates complement activation on self cells by possessing both cofactor activity for the Factor I mediated C3b cleavage, and decay accelerating activity against the alternative pathway C3 convertase, C3bBb. CFH protects self cells from complement activation but not bacteria/viruses. Due to the central role that CFH plays in the regulation of complement, there are many clinical implications arrising from aberrant CFH activity. Defects in CFH are the cause of complement factor H deficiency (CFH deficiency) and hemolytic-uremic syndrome (HUS). Genetic variation in CFH is associated with age-related macular degeneration type 4 (ARMD4) which is a multifactorial eye disease and the most common cause of irreversible vision loss in the developed world. |
Specificity : | Human Complement Factor H / CFH. No cross-reactivity with human cell lysate ( 293 cell line ) in ELISA. |
Immunogen : | Recombinant Human Complement Factor H / CFH Protein |
Host : | Mouse |
Isotype : | Mouse IgG1 |
Species : | Human |
Clone : | 9F6F3 |
Applications : | WB |
Dilution : | This antibody can be used at 1-2 μg/mL with the appropriate secondary reagents to detect Human CFH in WB. Using a DAB detection system, the detection limit for Human CFH is approximately 20 ng/lane under non-reducing conditions and 4 ng/lane under reducing conditions. |
Preparation : | This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, human cell-derived, recombinant Human Complement Factor H / CFH. The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography. |
Format : | 0.2 μm filtered solution in PBS with 5% trehalose |
Storage : | This antibody can be stored at 2-8 °C for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20 to -70°C. Preservative-Free.Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles. |
