Overview
Fibroblast growth factor receptor 1 (FGFR1), also known as basic fibroblast growth factor receptor 1, fms-related tyrosine kinase-2 / Pfeiffer syndrome, and CD331, is a receptor tyrosine kinase whose ligands are specific members of the fibroblast growth factor family. FGFR1 has been shown to be associated with Pfeiffer syndrome. -

What is fgfr1 protein?
FGFR1 protein stands for Fibroblast Growth Factor Receptor 1. It is a transmembrane receptor tyrosine kinase that is integral to many important biological processes:FGFR1 helps mediate the effects of fibroblast growth factors (FGFs), which are involved in processes like embryonic development, cell differentiation, cell proliferation, tumor growth, and angiogenesis.When FGFs bind to FGFR1, it causes FGFR1 to dimerize and autophosphorylate its cytoplasmic tyrosine kinase domains.This leads to activation of downstream signaling cascades like Ras-MAPK, PI3K-Akt, and PLCγ pathways.
FGFR1 is expressed in many tissues including epithelial, neuronal, endothelial cells during development and tumorigenesis.Gain-of-function mutations in FGFR1 can cause diseases by overactivating downstream signaling. Loss of function mutations also lead to diseases.FGFR1 play essential roles in development of tissues like bone, brain, lungs. It also contributes to wound healing, regeneration and various cancers when dysregulated.FGFR1 is a key cell surface receptor that mediates many critical FGF-induced biological functions through activation of intracellular signaling pathways upon ligand binding.
What is the function of fgfr1 protein?
Signal transduction: FGFR1 acts as a receptor for fibroblast growth factors (FGFs) on the cell surface. Binding of FGF ligands causes FGFR1 dimerization and autophosphorylation of its tyrosine kinase domains. This leads to activation of intracellular signaling cascades like Ras-MAPK, PI3K-Akt, PLCγ pathways.
Cell proliferation: FGFR1 signaling promotes proliferation of many cell types during development and tissue repair. It plays a role in regulating cell cycle progression, mitosis, and is crucial for growth and survival.
Embryonic development: FGFR1 signaling controls key processes like angiogenesis, limb development, bone formation, organogenesis etc. Important for development of tissues like lung, brain, bone during embryonic morphogenesis.
Fgfr1 related signaling pathway
RAS-MAPK pathway: Binding of FGF ligands leads to autophosphorylation and recruitment of FGFR substrate 2 (FRS2). This activates RAS, RAF, MEK and ERK kinases inducing gene expression changes influencing proliferation, differentiation and survival.
PI3K-Akt pathway: FRS2 also recruits and activates PI3K, leading to phosphorylation and activation of Akt. This pathway regulates cell growth, survival and metabolism.
PLCγ pathway: PLCγ is phosphorylated by FGFR1 inducing downstream PKC and calcium signaling regulating various cellular responses.
