Overview
The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based on its expression early in embryogenesis, the BMP encoded by this gene has a proposed role in early development. In addition, the fact that this BMP is closely related to BMP5 and BMP7 has led to speculation of possible bone inductive activity. -

Background
What is bmp8b protein?
BMPs are a group of signaling molecules that play important roles in bone formation and many other developmental processes. They signal through BMP receptors. BMP8B is classified in the BMP8 subfamily, along with BMP8A. BMP8A and BMP8B are closely related and their genes are located next to each other.
In mice, BMP8B is expressed early in development in the extraembryonic ectoderm, which signals to the proximal epiblast to induce primordial germ cell formation. As shown in this study, BMP8B is required for normal PGC generation. BMP8B also plays roles later in male germ cell development and spermatogenesis. Mice lacking functional BMP8B show defects in initiating and maintaining spermatogenesis.
The protein functions by binding to BMP receptors on target cells and activating intracellular SMAD signaling pathways, which regulate the expression of genes involved in cell differentiation and other processes.
What is the function of bmp8b protein?
Induction of primordial germ cell (PGC) formation: BMP8B is expressed in the extraembryonic ectoderm during early embryogenesis. It signals to proximal epiblast cells adjacent to the extraembryonic ectoderm to induce their specification into PGCs.
Requirement for normal PGC generation: Mice lacking functional BMP8B show severe reduction or absence of PGCs, indicating BMP8B is essential for proper PGC formation.
Role in extraembryonic mesoderm development: BMP8B knockout embryos show defects in allantois development, such as delayed initiation and shorter length. This suggests BMP8B also influences formation of extraembryonic tissues derived from mesoderm.
Bmp8b related signaling pathway
BMP8B signals by binding to type I and type II serine/threonine kinase receptors on the surface of target cells. The major receptors it signals through are: BMPR1A (also known as ALK3), BMPR1B (also known as ALK6), BMPR2.
When BMP8B binds these receptors, it induces phosphorylation of receptor-regulated SMAD (R-SMAD) proteins, specifically SMAD1, SMAD5, and SMAD8.
The phosphorylated R-SMADs then form a complex with the co-SMAD (SMAD4). This activated R-SMAD/co-SMAD complex translocates to the nucleus.
In the nucleus, the SMAD complex associates with other transcription factors to regulate the expression of target genes involved in various signaling responses, such as cell differentiation, proliferation and apoptosis.
Some of the key target genes of the BMP8B-SMAD pathway include ID proteins (inhibitors of differentiation), MSX proteins, and components of the Wnt signaling pathway like Lef1.
BMP8B Related Diseases
- Infertility in males: BMP8B knockout mice show defects in spermatogenesis and infertility. In humans, mutations in BMP8B have been linked to azoospermia (lack of sperm in semen) and oligospermia (low sperm count).
- Hypospadias: A congenital defect where the opening of the urethra is on the underside of the penis instead of at the tip. BMP8B mutations may cause hypospadias in rare cases.
- Cancer: Aberrant BMP signaling, including from BMP8B, has been implicated in certain cancers like prostate, breast and lung cancer. BMP8B expression levels can influence tumor growth and metastasis.
- Skeletal malformations: As BMPs regulate bone development, some skeletal abnormalities have been tied to BMP8B mutations, though rare. Conditions like acrocephalosyndactyly have been linked to BMP8B defects.
- Fertility treatment - Recombinant BMP8B protein could help treat male infertility caused by low BMP8B levels. It may promote spermatogenesis.
- Tissue engineering - BMP8B plays roles in bone formation. It could be used to help engineer bone grafts or stimulate bone/cartilage regeneration.
- Wound healing - Like other BMPs, BMP8B may aid healing of problematic wounds or burns by promoting cell growth and angiogenesis.
