During zebrafish embryogenesis, microRNA miR-430 contributes to restrict Nanos1 and TDRD7 to primordial germ cells (PGCs) by inducing mRNA deadenylation, mRNA degradation, and translational repression of nanos1 and tdrd7 mRNAs in somatic cells. The nanos1 and tdrd7 3'UTRs include cis-acting elements that allow activity in PGCs even in the presence of miRNA-mediated repression. Using a GFP reporter mRNA that was fused with tdrd7 3'UTR, a germline-specific RNA-binding protein DAZ-like (DAZL) can relieve the miR-430-mediated repression of tdrd7 mRNA by inducing poly(A) tail elongation (polyadenylation) in zebrafish. DAZL can also enhance protein synthesis via the 3'UTR of dazl mRNA, another germline mRNA targeted by miR-430. This study indicated that DAZL acts as an "anti-miRNA factor" during vertebrate germ cell development.

Fertility and gamete reserves are maintained by asymmetric divisions of the germline stem cells to produce new stem cells or daughters that differentiate as gametes. Before entering meiosis, differentiating germ cells (GCs) of sexual animals typically undergo cystogenesis. Here, researchers investigated cystogenesis in zebrafish and found that early GCs are connected by ring canals, and show that Deleted in azoospermia-like (Dazl), a conserved vertebrate RNA-binding protein (Rbp), is a regulator of this process. Analysis of dazl mutants revealed the essential role of Dazl in regulating incomplete cytokinesis, germline cyst formation and germline stem cell specification before the meiotic transition. Accordingly, dazl mutant GCs form defective ring canals, and ultimately remain as individual cells that fail to differentiate as meiocytes. In addition to promoting cystoblast divisions and meiotic entry, dazl is required for germline stem cell establishment and fertility.

The protein Dazl (Deleted in Azoospermia-like) is an important reproduction-related protein in zebra fish (Danio rerio). The following are some applications and research progress of Dazl protein in zebrafish:

Research on reproductive development: Dazl protein plays a key role in germ cell development of zebrafish. Studying the function of Dazl protein is helpful to understand the development process of germ cells and the pathogenesis of related diseases. Dazl protein plays an important role in spermatogenesis. By studying the expression and function of Dazl protein in zebrafish, we can reveal its specific role in sperm formation and provide potential targets for the treatment of male infertility.

Disease treatment research: Mutations or deletions of the Dazl gene in zebrafish can mimic certain human reproductive system disorders, such as azoospermia. Studying these models can help reveal the molecular mechanisms of these diseases and provide clues for developing treatments. Using zebrafish Dazl gene knockout model for drug screening and toxicity testing can evaluate the effects of drugs on reproductive system and provide important information for drug safety evaluation. By studying the response of Dazl proteins to environmental pollutants in zebrafish, the effects of these pollutants on reproductive health can be assessed.

Other basic research: Using zebrafish as a model organism, the function of Dazl protein was investigated by gene knockout technology. Dazl protein may be involved in epigenetic mechanisms regulating gene expression. Studying its epigenetic regulation in zebrafish will help to understand the complexity of gene expression regulation.
