Description : The histamine H1-receptor is expressed in numerous cell types and couples to Gq-proteins to activate phospholipase C. In Sf9 insect cells, rH1R couples to the endogenous insect cell Gq-protein. Co-expression with RGS proteins facilitates detection of high-affinity GTPase activity. rH1R reconstituted with RGS4 was prepared from Sf9 cells coinfected with rH1R- and RGS4-encoding baculoviruses. rH1R contains a N-terminal FLAG-tag and a C-terminal hexahistidine (His6)-tag for immunological detection, to allow purification and to provide additional protection against proteolysis.
