Description : | Histidine ammonia-lyase (or histidase, or histidinase) is an enzyme that in humans is encoded by the HAL gene. Histidase converts histidine into ammonia and urocanic acid. Histidine ammonia-lyase is a cytosolic enzyme catalyzing the first reaction in histidine catabolism, the nonoxidative deamination of L-histidine to trans-urocanic acid. |

Preparation : | HEK293T cells were transiently transfected with MegaTran Transfection Reagent and TrueORF cDNA plasmid. Transfected cells were cultured for 48-72 hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix, 1mM PMSF and 1mM Na3VO4), and then centrifuged to clarify the lysate. Recombinant proteins were purified through anti-DDK affinity columns. Protein concentration was measured by BCA kit. |
