Antigen Description : | ErbB2, also known as CD340 and HER2, is a type I membrane glycoprotein. It belongs to the epidermal growth factor (EGF) receptor family. ErbB2 cannot bind growth factors due to the lacking of ligand binding domain of its own and autoinhibited constitutively. However, this receptor forms a heterodimer with other ligand-bound EGF receptor family members, therefore stabilizes ligand binding and enhances kinase-mediated activation of downstream molecules. ErbB2 mediates signalling pathways which involve mitogen-activated protein kinase and phosphatidylinositol-3 kinase, this receptor plays an key role in development, cell proliferation and differentiation. ErbB2 is widely expressed in epithelial cells, and amplification and/or overexpression of the this gene has been reported associated with malignancy and a poor prognosis in numerous carcinomas, including breast, prostate and ovarian cancers. |
Specificity : | Human ErbB2 / HER2 / CD340 |
Immunogen : | Recombinant human ErbB2 protein |
Isotype : | Rabbit IgG |
Clone : | A512 |
Conjugation : | FITC |
Applications : | FCM; WB |
Dilution : | Western blot: This antibody can be used at 1-2 μg/mL with the appropriate secondary reagents to detect Human ErbB2 in WB. Using a DAB detection system, the detection limit for Human ErbB2 is approximately 1 ng/lane under non-reducing conditions. |
Preparation : | This antibody was obtained from a rabbit immunized with purified, human cell-derived, recombinant human ErbB2 / HER2 extracellular domain and conjugated with FITC under optimum conditions, the unreacted FITC was removed |
Format : | Aqueous solution containing 0.5% BSA and 0.1% sodium azide |
Concentration : | 5 μl/Test, 0.2 mg/ml |
Storage : | This antibody is stable for 12 months from date of receipt when stored at 2-8 °C. Protected from prolonged exposure to light. Do not freeze ! Sodium azide is toxic to cells and should be disposed of properly. Flush with large volumes of water during disposal. |

Ask a question1.Prepare the streptavidin-coated plate by washing it with PBS (phosphate-buffered saline) or another suitable buffer to remove any impurities. 2.Dilute the Avi-tagged protein in the desired buffer at the desired concentration. It is important to use a buffer that is compatible with the protein and does not interfere with its activity or stability. 3.Add the diluted protein to the streptavidin-coated plate and incubate it for a suitable amount of time at room temperature or 4°C. The exact conditions will depend on the specific protein and the desired level of immobilization. 4.Wash the plate with a suitable buffer to remove any unbound protein. 5.Block the remaining binding sites on the plate with a suitable blocking agent such as BSA (bovine serum albumin) or casein. 6.Wash the plate again to remove any unbound blocking agent. 7.Use the immobilized protein for further experiments such as ELISA, Western blotting, or other assays.
