Usage Protocol : • Allow the medium to warm to room temperature before use.
• For transfection, passage 293 cells in 1× CBM293 medium, with a recommended minimum of 3 passages for adaptation, in suspension culture in shaking flasks or bioreactors, with a seed cell passage density greater than 0.3×10^6 cells/mL. (Shaking bed speed reference: 150-180 rpm)
• Grow 293 seed cells to a density of 1.5-3.0×10^6 cells/mL, dilute the cell density to 0.8-1.0×10^6 cells/mL with 1× fresh CBM293 medium, and continue to culture the cells for 24 hours or less until the density reaches 1.5-3.0×10^6 cells/mL (not exceeding 3.0×10^6 cells/mL), then proceed with transfection.
• After 24 hours post-transfection, add supplement CBM-feed to the culture medium at a volume ratio of 2% (e.g., 2mL to 100mL), add once daily. Alternatively, add at 4% every two days.
• Typically, harvest and purify after 7 days post-transfection or when cell viability drops below 55%.
