Form : Lyophilized from sterile PBS, pH 7.4. Normally 5 % - 8 % trehalose, mannitol and 0.01% Tween80 are added as protectants before lyophilization.
Bio-activity : 1. Immobilized recombinant mouse CTLA4-His at 2 μg/ml (100 μl/well) can mouse CD80-Fc. The EC50 of mouse CD80-Fc is 1-9 ng/ml. 2. Immobilized recombinant mouse CTLA4-His at 10 μg/ml (100 μl/well) can human B7-1-Fc. The EC50 of human B7-1-Fc is 2-14 ng/ml. 3. Measured by its ability to inhibit IL-2 secretion by stimulated Jurkat human acute T cell leukemia cells. The ED50 for this effect is 0.1-0.5μg/mL when stimulated with 1 μg/mL Recombinant Human B7-1/CD80.
Molecular Mass : The recombinant mouse CTLA4 consists of 138 amino acids and has a predicted molecular mass of 15.46 kDa. In SDS-PAGE under reducing conditions, the apparent molecular mass of mouse CTLA4 is approximately 25-30 kDa due to glycosylation.
Endotoxin : < 1.0 EU per μg of the protein as determined by the LAL method.
Purity : > 95 % as determined by SDS-PAGE
Storage : Samples are stable for up to twelve months from date of receipt at -20°C to -80°C. Store it under sterile conditions at -20°C to -80°C. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles.
Reconstitution : It is recommended that sterile water be added to the vial to prepare a stock solution of 0.2 ug/ul. Centrifuge the vial at 4°C before opening to recover the entire contents.
