Description : MMEL1 is azinc protease of the neprilysin (NEP) family, which also includes NEP, ECE-2,PEX, DINE, Kell and several NEPlike proteins. It is encoded by the MMEL1 genein the human genome and is also known as soluble secreted endopeptidase (SEP),Neprilysinlike 1 and Neprilysin-2. Highly expressed in testis, the cDNApredicted a type II transmembrane protein with a short cytoplasmic tail and alarge ectodomain. Both membranebound and soluble forms were comparable withregard to model substrates, pH optima and inhibitor profiles. The ectodomainof human MMEL1 was expressedwith an N-terminal His tag and purified.
Form : Supplied as a 0.2 μmfiltered solution in Tris, NaCl and ZnCl2.
N-terminalSequence : His
MolecularWeight : 82 kDa
Activity : Measuredby its ability to cleave the fluorogenic peptide substrate, Mca-RPPGFSAFK(Dnp)-OH.The specific activity is > 250 pmoles/min/μg, as measured under thedescribed conditions.
Endotoxin Level : <1.0EU per 1μg of the protein by the LAL method.
SDS-PAGE : 99 kDa,reducing conditions.
Purity : >95%,by SDS-PAGE under reducing conditions and visualized by silver stain.
Storage : Use amanual defrost freezer and avoid repeated freezethaw cycles. 6 monthsfrom date of receipt, -20 to -70°C as supplied. 3 months,-20to-70°C under sterile conditions after opening.
