Overview
Fructose-1,6-bisphosphatase 1, a gluconeogenesis regulatory enzyme, catalyzes the hydrolysis of fructose 1,6-bisphosphate to fructose 6-phosphate and inorganic phosphate. Fructose-1,6-diphosphatase deficiency is associated with hypoglycemia and metabolic acidosis. -

What is FBP1 protein?
FBP1 (fructose-bisphosphatase 1) gene is a protein coding gene which situated on the long arm of chromosome 9 at locus 9q22. Fructose-1,6-bisphosphatase 1(FBP1) is a gluconeogenesis regulatory enzyme, catalyzes the hydrolysis of fructose 1,6-bisphosphate to fructose 6-phosphate and inorganic phosphate. The FBP1 protein is consisted of 338 amino acids and its molecular mass is approximately 36.8 kDa.
What is the function of FBP1 protein?
FBP1 appears to modulate glycerol gluconeogenesis in liver. It is an important regulator of appetite and adiposity. The expression of FBP1 in liver will increase after nutrient excess increases circulating satiety hormones and reduces appetite-stimulating neuropeptides and thus it seems to provide a feedback mechanism to limit weight gain.

FBP1 Related Signaling Pathway
The signaling pathways involved in FBP1 (fructose-1, 6-diphosphatase) are gluconeogenesis and glycolysis. In addition, the activity of FBP1 is regulated by many factors, including AMP, Insulin, PPARα, etc. PPARα is a nuclear receptor whose activation can inhibit FBP1 transcription. AMPK is an energy-sensitive kinase whose activity is regulated by changes in the intracellular AMP/ATP ratio. Activated AMPK can inhibit the expression and activity of FBP1, thereby reducing gluconeogenesis.
FBP1 Related Diseases
FBP1 is mainly involved in the metabolic process of glucose. If the function of this enzyme is abnormal, or the expression of FBP1 is low, it is associated with hypoglycemia and metabolic acidosis. And it may affect the normal metabolism of glucose, which may lead to a series of metabolic diseases, such as diabetes and obesity. Other related diseases include glycogen storage disease type I (GSD-I), hepatic insulin resistance and hepatocellular carcinoma (HCC).
