FABP1

  • Official Full Name

fatty acid binding protein 1, liver
  • Overview

This gene encodes the fatty acid binding protein found in liver. Fatty acid binding proteins are a family of small, highly conserved, cytoplasmic proteins that bind long-chain fatty acids and other hydrophobic ligands. This protein and FABP6 (the ileal fatty acid binding protein) are also able to bind bile acids. It is thought that FABPs roles include fatty acid uptake, transport, and metabolism. Background

    What is fabp1 protein?

FABP1 is predominantly expressed in the small intestine and colon epithelial cells. It plays a role in fatty acid absorption from the diet.FABP1 binds saturated and unsaturated long-chain fatty acids inside cytoplasm and delivers them to various organelles like endoplasmic reticulum, mitochondria and nuclei. This facilitates fatty acid metabolism. Due to its role in fatty acid uptake and transportation, FABP1 is important for dietary fat digestion/absorption as well as energy production through beta-oxidation of fatty acids. It comprises 132 amino acids folded into a beta-barrel structure that creates a hydrophobic cavity to encapsulate fatty acid molecules.

What is the function of fabp1 protein?

Fatty acid uptake and transport: FABP1 binds long-chain fatty acids inside intestinal and colon epithelial cells after their release from triacylglycerols during digestion. It transports the fatty acids to various intracellular sites involved in metabolism.

Fatty acid metabolism: FABP1 delivers fatty acids to organelles like mitochondria and peroxisomes for beta-oxidation and energy production. It transfers fatty acids to ER for synthesis of phospholipids, triglycerides and cholesterol.

Bile acid absorption: FABP1 facilitates the absorption of dietary bile acids which are important for fat digestion and absorption.

Gene regulation: It may act as a lipid signal and regulate expression of genes involved in fat absorption and metabolism.

    Fabp1 related signaling pathway

PPAR signaling pathway: PPARγ regulates Fabp1 expression. FABP1 transports fatty acids that are ligands for PPARγ/α/δ, activating this vital lipid-sensing pathway.

Insulin signaling pathway: FABP1 facilitates fatty acid uptake and metabolism which impacts insulin sensitivity and glucose homeostasis via cross-talk with insulin signaling.

Wnt/β-catenin pathway: Intestinal FABP1 is regulated by transcription factor TCF4 downstream of the Wnt cascade, influencing absorption and metabolic genes.

Nrf2-mediated oxidative stress response: FABP1 binds oxidative lipid species transporting them for detoxification by antioxidant pathways involving Nrf2 transcription factor.

MAPK pathway: Extracellular signal-regulated kinase (ERK) pathway mediates FABP1 induction in response to fatty acids through an unclear mechanism.

    FABP1 Related Diseases

Obesity: Higher levels linked to obesity, adiposity and metabolic dysfunction.

Insulin resistance: Role in impaired fatty acid utilization contributing to diabetes.

Colorectal cancer: Overexpression promotes tumorigenesis through oncogenic signaling.

Inflammatory bowel disease: Upregulation in conditions like Crohn's disease and ulcerative colitis.

Non-alcoholic fatty liver disease: Aberrant hepatic FABP1 implicated in NAFLD pathogenesis.

    Biomedical Application of FABP1 Protein

    •  Cancer therapy: Targeting aberrant intestinal FABP1 signaling treats colorectal tumors.
    •  Obesity research: Regulation of its expression influences adiposity and metabolic syndrome.
    •  Drug development: Screening assays identify pharmacological FABP1 inhibitors for metabolic diseases.
