  • Overview

    Mediates pre-mRNA alternative splicing regulation. Binds to splice sites in pre-mRNA and regulates splice site selection. Binds and stabilizes cytoplasmic mRNA. Contributes to the regulation of translation by modulating the interaction between the mRNA and eukaryotic initiation factors (By similarity). Regulates the transcription of numerous genes. Its transcriptional activity on the multidrug resistance gene MDR1 is enhanced in presence of the APEX1 acetylated form at 'Lys-6' and 'Lys-7'. Binds to promoters that contain a Y-box (5'-CTGATTGGCCAA-3'), such as MDR1 and HLA class II genes. Promotes separation of DNA strands that contain mismatches or are modified by cisplatin. Has endonucleolytic activity and can introduce nicks or breaks into double-stranded DNA (in vitro). May play a role in DNA repair. Component of the CRD-mediated complex that promotes MYC mRNA stability.<br/>The secreted form acts as an extracellular mitogen and stimulates cell migration and proliferation.

    Case Study 1: Lu K. et al. Liver Int. 2024

    Metabolic dysfunction-associated steatotic liver disease (MASLD) leads in chronic liver issues, but its mechanisms remain vague. We found a lncRNA called LNCHC, studying its role in MASLD. LNCHC binds with YBX1, preventing its breakdown and stabilizing PNPLA3 mRNA, which cuts liver fat. Though LNCHC, YBX1, and PNPLA3 help control lipid buildup, they may also worsen metabolic dysfunction-related liver inflammation.

    Case Study 2: Chiba M. et al. Int J Mol Sci. 2023

    Aging cells release inflammatory proteins and small vesicles called SASP, which contribute to age-related diseases. They produce SATII RNA, ramping up inflammatory genes and moving in vesicles to other cells. We found YBX1 helps SATII RNA get into these vesicles. Cells receiving these vesicles age faster and express more inflammatory genes. Cutting YBX1 levels reduces SATII RNA in vesicles and limits SASP spread. Higher YBX1 is found in cancerous tissues and is linked to worse outcomes in breast and ovarian cancers.
