Overview
The protein encoded by this gene is a member of the cyclin-dependent protein kinase (CDK) family. CDK family members are highly similar to the gene products of S. cerevisiae cdc28, and S. pombe cdc2, and known as important cell cycle regulators. This kinase was found to be a component of the multiprotein complex TAK/P-TEFb, which is an elongation factor for RNA polymerase II-directed transcription and functions by phosphorylating the C-terminal domain of the largest subunit of RNA polymerase II. This protein forms a complex with and is regulated by its regulatory subunit cyclin T or cyclin K. HIV-1 Tat protein was found to interact with this protein and cyclin T, which suggested a possible involvement of this protein in AIDS. [provided by RefSeq, Jul 2008] -

What is CDK9 protein?
CDK9 (cyclin dependent kinase 9) gene is a protein coding gene which situated on the long arm of chromosome 9 at locus 9q34. The protein encoded by this gene is a member of the cyclin-dependent protein kinase (CDK) family. CDK family members are highly similar to the gene products of S. cerevisiae cdc28, and S. pombe cdc2, and known as important cell cycle regulators. The CDK9 protein is consisted of 372 amino acids and its molecular mass is approximately 42.8 kDa.
What is the function of CDK9 protein?
CDK9 protein was found to be a component of the multiprotein complex TAK/P-TEFb, which is an elongation factor for RNA polymerase II-directed transcription and functions by phosphorylating the C-terminal domain of the largest subunit of RNA polymerase II. This protein forms a complex with and is regulated by its regulatory subunit cyclin T or cyclin K. Phosphorylation of CDK9 regulates DNA damage repair and apoptosis signaling pathways. CDK9 also interacts with proteins related to apoptosis and cell cycle regulation, and is involved in the regulation of cell proliferation and cell fate determination.

CDK9 Related Signaling Pathway
CDK9 is mainly involved in the process of transcription extension, and can promote gene expression through the kinase activity of CDK9/P-TEFb complex. In addition, CDK9 is involved in cell cycle regulation. When cells enter the S and G2 phases, CDK9 is activated and can promote the cell to enter mitosis.
CDK9 Related Diseases
Diseases associated with CDK9 include Hypothyroidism, Congenital, Nongoitrous, 6 and Nut Midline Carcinoma. Due to its key role in transcriptional regulation, abnormal activity of CDK9 has been linked to a variety of diseases, including certain types of cancer, cardiovascular disease, and viral infections such as HIV.
Bioapplications of CDK9
Because of its potential role in a variety of diseases, CDK9 has become an attractive target for drug development. For HIV treatment in particular, drugs that target CDK9 may help inhibit viral replication.
