  • Overview

    The tuberous sclerosis 1 protein interacts with TSC2 (tuberin) to form a functional tumor suppressor complex that regulates cell proliferation and growth. Tuberous sclerosis is an autosomal dominant tumor suppressor gene syndrome due to inactivating mutations in either TSC1 or TSC. The TSC1/TSC2 complex functions as a negative regulator of mTOR activity and inhibits 70S6K and 4EBP1 phosphorylation in cells. The mechanism of inhibition is to due in part to intrinsic GAP activity acting upon the small GTPase, Rheb. The PI-3K pathway inhibits TSC1/TSC2 via Akt phosphorylation and subsequent destabilization of the functional TSC1/TSC2 complex. Activation of the complex occurs via AMPK,Which senses stress and energy levels in the cell. Phosphorylation of the TSC1/TSC2 complex by AMPK results in activation of the complex and inhibition of mTOR activity.
