DNA methylation cannot be directly investigated using conventional DNA amplification because methylation markers cannot be maintained during this period. Bisulfite conversion is one of the most widely used methods to convert sample with DNA methylation markers into appropriate amplification and downstream analysis templates. Creative BioMart is capable to provide our customers with superior bisulfite conversion service, ensuring high-quality templates of DNA methylation analysis for downstream applications based on bisulfite conversion.
Basic Principle of Bisulfite Conversion
The principle of bisulfite conversion is based on the chemical conversion of unmethylated cytosine to uracil, while methylated cytosine is exempt from this conversion, allowing DNA methylation to be determined at the single-base resolution. Bisulfite conversion is a process in which DNA is denatured (made into a single strand) and treated with sodium bisulfite, resulting in deamination of unmethylated cytosine to uracil, while methylated cytosine (5-mC and 5-hmC) remains unchanged.
