Description: | Protein arginine methylation is a post-translational modification that contributes to numerous biological functions such as cell signaling and transcriptional repression. In mammals arginine methylation is catalyzed by three classes of protein arginine methyltransferase (PRMT) enzymes. Types I-III catalyze the transfer of a methyl group from S-adenosyl-L-methionine to the terminal (ω) guanidino nitrogen of arginine residues to produce a monomethylarginine. Type I PRMTs further methylate the ω-nitrogen to produce an asymmetric dimethylarginine while Type II PRMTs produce symmetric dimethylarginine. PRMT8 is a type I methyltransferase enzyme shown to methylate the glycine-arginine rich region in fibrillarin. PRMT8 shares approximately 80% amino acid similarity with PRMT1. Unlike PRMT1, which is ubiquitously expressed, PRMT8 has limited tissue distribution. Northern blot analysis of various tissues suggest that PRMT8 mRNA expression is limited to the brain. PRMT8 resides on the plasma membrane and this subcellular localization is mediated by a myristoylated glycine residue and a patch of basic amino acids at the N-terminal end of the protein. The N-terminal domain of PRMT8 is believed to have a regulatory function as truncation of the first 60 amino acids has been shown to increase enzymatic activity. |
