Description: | HDAC5 is a member of the histone deacetylase family. It is responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4). Histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Histone deacetylases act via the formation of large multiprotein complexes. HDAC5 possesses histone deacetylase activity and represses transcription when tethered to a promoter. It coimmunoprecipitates only with HDAC3 family member and might form multicomplex proteins. It also interacts with myocyte enhancer factor-2 (MEF2) proteins, resulting in repression of MEF2-dependent genes. |

Activity: | > 10000 pmol/min/μg. Assay condition: 50 mM Tris-HCl, pH8.0, 136 mM NaCl, 2.8 mM KCl, 1 mM MgCl2, 0.5 mg/ml BSA, 0.005% Tween-20, 20 M AMC-RHKKAc and HDAC5. HDAC reaction was done for 30 min at 37 centigrade followed by treatment with equivoluminal HDAC developer buffer (40 μM Trypsin, 40 μM CaCl2 and 20 μM Trichostatin A for ~60 min at RT. |
