| Scientific Background: | The CDX family members (CDX1, CDX2, and CDX4) are Caudal-related transcription factors implicated in anterior-posterior patterning during embryonic development, regulation of cardiac differentiation in stem cells and down-regulation of genes associated with neuronal growth and vascular endothelial growth. The caudal-type homeobox 1 (CDX1) transcription factor controls enterocyte differentiation in the colon and is also central to the capacity of a colorectal cancer (CRC) cell line to differentiate. CDX1 is a negative marker of CRC stem cells. The expression of CDX1 is itself regulated by methylation at the CDX1 promoter region, and its expression is conversely correlated with expression of the polycomb complex protein BMI-1, which is involved in maintenance of quiescent stem cells and is expressed in cancer stem cells. |
