| Scientific Background: | PML (Promyelocytic Leukemia) protein is a transcription factor originally identified due to a chromosomal aberration fusing it to the Retinoic Acid Receptor alpha (RAR-A) in cases of Acute Promyelocytic Leukemia (APL). Besides transcriptional regulation, PML is involved in many processes, including the response to DNA damage, cell cycle regulation, viral infection response and apoptosis. These functions of PML involve the localization of PML to unique sub-nuclear structures called PML Nuclear Bodies (PML NBs). Over fifty proteins are know to be localized to PML NBs, either transiently or constitutively. These Nuclear Bodies are disrupted in PML knockout mice. PML is subject to a number of post-translational modifications, including phosphorylation, ubiquitylation and sumoylation. |
