Scientific Background: | With a critical role in long-term transcriptional silencing, DNA methylation represents a key layer of epigenetic information which is essential for embryonic development, germline differentiation, functional maturation of neuronal tissues etc. DNA-methyltransferases such as DNMT3A, DNMT3B and DNMT3L are unique enzymes with the ability to transfer methyl groups derived from folate intermediates to nucleic acids. DNA (cytosine-5)-methyltransferase 3A or DNMT3A is required for genome-wide de novo methylation and for establishment of DNA methylation patterns during development. Localized in the nucleus and cytoplasm, it accumulates in major satellite repeats at pericentric heterochromatin and is activated through binding to its regulatory factor DNMT3L. DNA methylation is coordinated with methylation of histones and DNMT3A modifies DNA in a non-processive manner, and it also methylates non-CpG sites. DNMT3A preferentially methylates DNA linker between 2 nucleosomal cores and is inhibited by histone H1. It plays a role in paternal/maternal imprinting and is required for methylation of most imprinted loci in germ cells. DNMT3A acts as a transcriptional corepressor for ZNF238 and actively represses transcription through HDAC. |
