Background: | While the other ten human HDACs fall into either the class I (HDACs 1-3, 8) or class II (HDACs 4-7, 9, 10) homology groups, HDAC11 is the sole human representative in class IV.In contrast to the ubiquitous expression of most human HDACs, high level expression of HDAC11 is limited, in normal human tissues, to brain, heart, skeletal muscle, kidney and testis, suggesting possible tissue-specific or developmental functions. HDAC11 appears to play a role in neural cell maturation, particularly that of oligodendrocytes. HDAC11 deacetylation of histone H3 lysines 9 and 14 in the myelin basic protein and proteolipid protein genes stimulates their expression in oligodendrocytes, a notable example in which histone deacetylation, frequently associated with gene silencing, may in some cases have the opposite effect. HDAC11 negatively regulates IL-10 expression in antigen presenting cells (APCs), thus influencing whether APCs induce T cell activation or tolerance. Elevated HDAC11 expression is seen in a number of cancer cell lines. In myeloid leukemia lines, several HDAC inhibitors strongly induce HDAC11 expression, although the induction is more rapid with some inhibitors (TSA, SAHA) than others (butyrate, valproate). Such effects may be important to consider in the development and clinical application of HDAC inhibitors as cancer therapeutics. |
