Description:  SUV420H1 and SUV420H2 are SET domain containing methyltransferases responsible for di- and trimethylation of histone H4 lysine 20 (H4K20me2 and H4K20me3) at pericentric heterochromatin. Monomethylation of histone H4K20 is specifically catalyzed by SET8 in higher eukaryotes. H4K20 dimethylation is believed to be required for proper DNA damage response, through recruitment of 53BP1. Trimethylation of the H4K20 mark leads to a repressive chromatin state corresponding to a suppression of position-effect variegation (PEV) at constitutive heterochromatin. Data obtained from murine embryonic fibroblasts deficient in SUV20H1 and SUV20H2 show increased frequencies of teleomere recombination suggesting SUV420H methyltransferases play an important role in maintaining telomere structure.
