Description:  The nuclear receptor SET domain-containing (NSD) family of histone lysine methyltransferases are important enzymes responsible for maintaining chromatin integrity. The NSD family consists of three members NSD1, NSD2, and NSD3. Perturbations in the NSD family can lead to several diseases such as Sotos syndrome, Wolf-Hirschorn syndrome, and cancers such as acute myeloid leukemia (AML) and breast cancer. NSD1, which can methylate histone H3 at lysine 36 (H3K36) and H4K20, is a large protein with 2,696 residues. It has several motifs near the C-terminal region that have been shown to be essential for chromatin regulation. Furthermore, NSD1 contains a catalytic lysine methyltransferase SET domain and four zinc-binding PHD fingers. NSD1 gene translocations, which promote expression of oncogenic NSD1 fusion proteins, are frequently found in patients with AML and breast cancer. In contrast, inactivation of NSD1 in neuroblastomas and glioblastomas has been shown to result in tumor suppression.
