| Description: | The nuclear receptor SET domain-containing (NSD) family of histone lysine methyltransferases are important enzymes responsible for maintaining chromatin integrity. The NSD family consists of three members NSD1, NSD2 (WHSC1, MMSET), and NSD3 (WHSC1-like). NSD2 has been implicated in multiple myeloma, an incurable malignancy in mature plasma cells, being involved in recurrrent t(4;14) translocations with the immunoglobulin promotor/enhancer. NSD2 is responsible for the post-translational modification of histones H3 and H4. The methylation target of NSD2 is dependent on the nature of the substrate. NSD2 specifically methylates H3K36 when provided with nucleosomal substrates, however, H4K44 is the primary target when native or recombinant octamers are used as substrates. In addition, H4K20 dimethylation catalyzed by NSD2 has been reported to increase locally at sites of DNA damage. This was proposed to be the trigger for the recruitment of 53BP1, an important mediator of the DNA damage response. Although, other sites on histones H3 and H4 have been reported H3K36 dimethylation may be the primary product of NSD2. |
