Description:  SETD2 is a histone methyltransferase that catalyzes the trimethylation of Histone H3 on lysine 36 (H3K36). SETD2 knockdown in mouse fibroblast cells demonstrates unaltered mono- and di-methylation but an impairment of trimethylation at H3K36 suggesting primarily trimethylase activity. However, it remains to be elucidated whether this methyltransferase activity is processive or non-processive. The WW domain of SETD2 has been shown to interact with hyperphosphorylated RNA polymerase II, indicating a broad role in transcriptional regulation. Evidence has also been found indicating SETD2 can automethylate itself, possibly influencing its methyltransferase activity. SETD2 interacts with huntingtin, the protein implicated in Huntington’s Disease, and may contribute to the progression of the disease.
