Description: | The acetylation of histone lysine residues plays a crucial role in the epigenetic regulation of gene transcription. Acetylated lysine residues are recognized by a small protein domain known as a bromodomain. Plant homeodomain (PHD) fingers are zinc binding domains comprised of ~50 amino acids and have a Cys4-His-Cys3 motif. PHD fingers are found in more than 100 nuclear proteins that play a role in regulating chromatin. Both the PHD and bromodomain in many proteins often work together to recognize post-translational modifications. TRIM28 (KAP-1) is a KRAB associated protein that is part of the tripartite motif family, which includes a RING, B1, B2, coiled-coil motif, a PHD domain and a bromodomain domain. The PHD finger of TRIM28 plays an important role in gene silencing. TRIM28 has been shown to interacts with SETD1 and other methyltransferases for gene silencing of endogenous retroviruses during embryogenesis. The PHD domain and bromodomain of TRIM28 have been shown to work together to facilitate lysine SUMOylation, which is a requirement for TRIM28 activity in gene silencing. |
