Significance: Soil urease catalyzes the hydrolysis of urea into ammonia and carbon dioxide. Its activity is positively correlated with soil microbial abundance, organic matter content, total nitrogen, and available nitrogen, serving as an indicator of soil nitrogen status.
Principle: Using urea as the substrate, ammonia nitrogen (NH3-N) produced by urease is quantified by the indophenol blue colorimetric method. The intensity of the blue color is proportional to ammonia concentration.
