Significance: Soil proteases participate in the transformation of amino acids, proteins, and other organic nitrogen-containing compounds in soil. Their hydrolysis products serve as nitrogen sources for higher plants. Soil neutral protease (S-NPT) catalyzes protein hydrolysis under neutral conditions and is closely related to soil organic matter, nitrogen content, and other soil properties.
Principle: Under neutral conditions, S-NPT hydrolyzes casein to produce tyrosine. Under alkaline conditions, tyrosine reduces phosphomolybdic acid to form tungsten blue, which exhibits a characteristic absorbance peak at 680 nm.
