Alkaline Phosphatase activity determination method and detection principle

At present, the common methods for detecting Alkaline Phosphatase activity in the human body are based on the principle of enzyme reactions. When Alkaline Phosphatase activity is present, the hydrolysis reaction of the substrate can be observed by adding a substrate, resulting in the formation of a yellow product. After reasonable color comparison, we can determine the content and activity of Alkaline Phosphatase based on the color difference generated by the sample.

The pNPP method is used to determine ALP activity through the enzymatic hydrolysis reaction of hydroquinone diphosphate (pNPP). The principle of this method is that pNPP is hydrolyzed into yellow phosphate under the catalysis of ALP, and its optical density can be measured by a spectrophotometer. This method is simple, easy to operate, with high accuracy and good sensitivity, and has been widely used in clinical testing.

The AS-5 rule is a method for determining ALP activity based on phosphate ion selective electrodes. Its principle is to add magnesium ions to the reaction system, causing ALP to catalyze the hydrolysis of substrate isopropyl phosphate (IAP) into orthophosphate, releasing phosphate ions, and causing electrical signals to appear on the electrode. ALP activity is measured by detecting the size of electrical signals. This method can avoid errors caused by different substrates and is also suitable for determining samples with acid phosphatase activity.
