ALDH, also known as aldehyde dehydrogenase, is one of the important metabolic enzymes in cells and plays an important role in oxidative metabolism. ALDH has the ability to oxidize aldehydes and is involved in many physiological and pharmacological processes, such as alcohol metabolism, drug metabolism, and chemical metabolism. In addition, research has found that ALDH can also regulate the proliferation and apoptosis of tumor cells.
The biological activity of ALDH can be attributed to its metabolic effect on aldehydes. ALDH represents cellular metabolism in an oxidative state, and its functional diversity is crucial for physiological and pharmacological research. They convert aldehydes into corresponding acid compounds through oxidation, which can consume the substrate of the redox process as a direct reaction product, while also producing appropriate chemical energy and metabolic products. Therefore, ALDH plays an important physiological role in metabolism and cellular signal transduction pathways.
The activity determination methods of ALDH mainly include chemical colorimetry, fluorescence method, luminescence method, etc. Fluorescence method is commonly used to determine the activity of ALDH. Its principle is to add the substrate of ALDH, acetone, to the determination mixture and combine it with a fluorescent probe targeting acetone. The fluorescence signal weakens with the conversion of acetone. This method can sensitively detect the activity of ALDH in trace samples. Chemical colorimetry is an enzymatic detection method established based on the spectral characteristics of NADH complexes. The principle is to add ALDH substrate to the determination mixture, combine with NAD dependent enzymes to form NADH, and then oxidize NADH and reduce certain chemical agents through a reaction to produce visible light absorption. Therefore, enzyme activity can be analyzed through light absorption values.
