Significance of Measurement: Acetyl-CoA is a central metabolic intermediate widely present in animals, plants, microorganisms, and cultured cells. It serves as a metabolic hub linking carbohydrate, lipid, and protein metabolism through the tricarboxylic acid cycle and oxidative phosphorylation. Acetyl-CoA is also a precursor for fatty acids, ketone bodies, cholesterol, and other bioactive molecules.
Principle of Measurement: Malate dehydrogenase catalyzes the conversion of malate to oxaloacetate with NADH production. Citrate synthase then catalyzes the condensation of acetyl-CoA and oxaloacetate to form citrate. The rate of NADH generation is proportional to acetyl-CoA content and is measured at 340 nm.
